When manufacturing biologics, contamination from mycoplasma and spiroplasma can contribute to a potential direct or indirect cause of human disease. Sources of contamination include culture medium components such as serum or trypsin, or even the laboratory personnel themselves. Testing for mycoplasma must be performed at various phases of product development, including raw materials, cell banks, viral seed stocks, unprocessed bulk harvest materials, and final products. Spiroplasma testing is also suggested for biopharmaceutical products that have been in contact with plant or insect materials.
Compendial mycoplasma testing procedures are culture-based and require 28 days of incubation. The lengthy turnaround is not amenable for rapid lot release testing for short-shelf life products such as cell and gene therapy products or for rapid in-process screening to detect potential contamination events.
